I have a camera link to the CLIPPER card through ETHERNET with MODBUS.
I still want to use PEWIN32 communicate to the CLIPPER through the ETHERNET .
Can I use a router as below[refer to the attachment]?

Yes. However, please keep in mind that the Modbus and Ethernet communication to PC are using the same resources and this may limit the bandwidth for one side if the other process has high demand for bandwidth.
